The Conversion Failure
Yesterday evening, I was really in the state of trembling because I was going to perform the conversion of the total RNA into cDNA. The last time I did was a one time success but I was not sure for today. I came early in the morning to the lab and proceed with the experiment for one hour. Then, I immediately conducted PCR experiment to test whether or not the cDNA could work. After 6 hours of patiently waiting, I visualized the amplicons on the gel and it turned out the gel was completely blank!..Only a trace of primer dimer could be seen. How frustrated I was that I finally went back home in vain....
