One Vector to Rule them All

Yesterday, I was really wrapped up with tones of labwork. The experiment was the important highlight of the quest for MaSp where I managed to insert the purified MaSp amplicon that was amplified from cDNA. Of course, I screened the ligation process whether or not it was successful and it was! Then, I let the pEasy cloning vector to be taken by the Trans 1 T1 phage cells by heat shock induction and incubated them overnight. What I found this morning, there were lota of colonies which have been transformed with the vector inserts! I will do the PCR screening tomorrow to see which colonies are positively transformed.. Crossing fingers to this! 

 You can see the white dots are the transformed bacteria with my vector inserts! Wait till I check on you guys..

The vector is positively ligated with the MaSp 12kb insert though not really have sharp band due to excess of DNA during pcr..
