The Long Lost of Correct cDNA Concentration

Yesterday, again I performed the amplification of MaSp using the diluted 1:10 cDNA, in a hope that MaSp band would emerge. I tested at different volume of cDNA with estimated concentrations. After 6 hours of waiting, the band was not yet visible. I am too tired of doing this LD-PCR using cDNA for MaSp amplification. I just could not replicate the right concentration of cDNA like the last time where I managed to obtain the MaSp band. Where are you cDNA, the appropriate cDNA concentration?..
