Where are you MaSpCL??
This morning, I had to subculture the C2 T1T1P cells to extract the pMaSpCl again. This was due to the inability of the sequencing process to capture the sequence from the gel cut sample. I was informed that the sample did not contain amy sequence of the insert instead only the vector and a chunk of E. coli's gDNA were obtained. I suspected that the ge cut sample still had the uncut pMaSpCl. But, I need to confirm this by exposing the plasmid to RE to a prolonged incubation time. Crossing fingers to this tomorrow!
