Putting Theory into Practical

Yesterday I performed the in vitro cleavage of NT using the sgRNAs that I designed and synthesized. I was really excited to apply everything I had learnt previously about CRISPR Cas9 after attending the two workshops conducted by Dr. Kumita from IPPT, USM. The process took only an hour 20 minutes to complete and the reactions were visualized on gel agarose. It was really amazing as the sgRNAs did make targeted cuts on the NT. But still, I need to optimize a bit with the concentration ratio etc. to get a better result. Yosh!
